چکیده

The effects of two beta-adrenergic blocking agents, pronethalol and propranolol, on ventricular fibrillation thresholds were tested in dogs. Pronethalol (4.0 mg/kg, 5 dogs) produced a mean increase of 45% in the fibrillation threshold. Propranolol (0.4 mg/kg, 7 dogs) failed to produce a significant change of the fibrillation threshold. To examine the possibility that the difference between the responses to pronethalol and propranolol might be related to the difference in dose of the beta-blocking agents, 4.0 mg/kg of propranolol was given to 3 dogs; this dose also failed to produce a consistent change of ventricular fibrillation threshold. These data indicate that under the conditions of our experiments it was possible to block the betaadrenergic receptors with propranolol without producing a change of the ventricular fibrillation threshold. The increase of the fibrillation threshold produced by pronethalol was probably a consequence of its quinidine-like properties rather than of its beta-receptor blockade. ADDITIONAL

The role of cyclic adenosine monophosphate in adrenergic effects on ventricular vulnerability to fibrillation in the isolated perfused rat heart.

The relation between myocardial tissue cyclic AMP (cAMP) and the vulnerability to ventricular fibrillation was assessed in the isolated perfused rat heart by measurement of ventricular fibrillation threshold (VFT) and vulnerable period duration (VP). Exogenous dibutyryl cyclic AMP (DBcAMP) reduced VFT and increased VP by a concentration-related action whereas exogenous cAMP did not. Theophyllin...

Roles of adrenergic and cholinergic stimulation in spontaneous atrial fibrillation in dogs.

OBJECTIVES We studied the effects of beta-adrenergic and cholinergic stimulation and blockade on spontaneous atrial fibrillation (AF) in the intact dog heart. BACKGROUND Paroxysmal AF is often preceded by changes in autonomic tone, but the relative roles of adrenergic and cholinergic influences on AF induction are not well known. METHODS Perfusion of catecholamines and acetylcholine (ACh), ...

Muscarinic effects of vagosympathetic trunk stimulation on the repetitive extrasystole (RE) threshold.

The precise mechanism of the vagus in opposing adrenergic influences on cardiac vulnerability has not been defined. In the present investigation, cholinergic stimulation was produced by administering the selective muscarinic agent methacholine (MCh) and ventricular vulnerability was assessed by measuring the repetitive extrasystole (RE) threshold. MCh produced a sustained 98 +/- 12% (P less tha...

Beta-Adrenergic Blockade Therapy for Autonomic Dysfunction is Less Effective for Elderly Patients with Heart Failure and Reduced Left Ventricular Ejection Fraction

OBJECTIVE Heart rate variability (HRV) has been reported to be an independent predictor of all-cause and sudden cardiac death in patients with heart failure. In the aging heart, however, both autonomic and cardiac functions appear to be altered. We assessed the relationship between aging and responsiveness of HRV and ventricular remodeling to beta-adrenergic blockade therapy in patients with he...
